He's a member of the underground faction of ANBU called Root. The Root's mission is to rule Konoha one day. Sai's missions in the past is to eliminate the possible oppositions to their plans from other villages. These range from top ninjas of other villages to S-Rank criminals. Root's leader, Danzou, sent Sai to Team Kakashi so that once Team Kakashi finds Sasuke, Sai can kill him. His mission is to kill Sasuke because Sasuke is among the list of the ninjas that Root must destroy. It's in the manga.

I'm sorry for using the term mission. I think the proper term is goal. It's Root's goal to rule Konoha one day. From your question, I assume you do not read the manga. Danzou is the rival of Sarutobi in becoming Hokage. But because Danzou is more of a military dictator in thinking, he was not chosen to succeed Nidaime (Second). After this, he formed Root and used harsh military discipline as training. In Root, there is no name, no emotion, only the mission.

Sai is one of Denzou's talented subordinates in the Root Division.
His real mission is to kill Sasuke which is viewed by Denzou as a threat to Konoha. He intends to kill Sasuke for the purpose of denying Orochimaru the chance to transfer himself to Sasuke's body and further destroy Konoha which is part of the plan. If Orochimaru cannot transfer from a strong body like that of Sasuke, then, Denzou wouldn't feel threatened with Orochimaru overtaking Konoha and wouldn't have a hard time killing Orochimaru himself since Denzou also love to rule the Konoha village.
