What Do You Look For When You Take A Role?

I'm working on this screenplay, and I see the character's in my head, but it would help to know what draws you to a character, what makes them real for you? So far I've only been an extra in a movie. I plan to do more in the future, but writing in where my focus is right now. So, tell me what should I stay away from? What's been done to death? What's your dream role?
