Should we use medicine like pain killer to get immdiate relief from back pain??

I am a software engineer and usually suffered by back pain when I sit on the computer chair for long hours. My friends suggested to take rest and try to do some exercise .
But It takes a week to recover from the pain . Can't I go for a medicine ...??
What harm is in this ??
